The only two seed I ever recall losing in the first round was South Carolina. They were knocked off by Coppin State I believe.

Richmond beat Syracuse in 1991, Santa Clara beat Arizona in '93, Coppin State beat South Carolina in '97 and Hampton beat Iowa State in 2001
